INTRODUCTION: The quality of work and skill performance requires knowledge about different operation modes of the electron microscope and proper preparation of samples. It is in this regard that the electron microscope should properly aligned, astigmatism free as well as considered different operation mode. The microstructure at highest levels of resolution is the goal of this workshop. Also an interpretation of pathogenic changes compared with poorly fixed samples should be taken into consideration. Of particular interest in this workshop is to prepare many samples to study its internal structure. Preparation includes different tissues of normal mice and bacteria.
Course Objective: The intended audience is PhD students and researchers with expertise/interest in their field of research using electron microscope. Therefore this workshop aims to train participants to be familiar with the basic parameters, which influence the image. Topics covered include the illumination system, electron lenses and their aberrations, image formation and resolution, the vacuum system and contamination. This course also aims to train participants to be familiar with the basic parameters, which affect sample preparations.
The course outline: In addition to practical sessions, this workshop includes the following lectures: Basic concepts of electron microscopy. Electron microscopy and cell biology. Sample preparation techniques. Deception by technical artifacts during interpretation of TEM micrographs. Ultrastructure of bacteria.
